SCNP identifies CLL donor subgroups by profiling functional immune signaling in the context of immune modulatory receptor expression

چکیده
Background Antibody therapeutics targeting the Immune Modulatory Receptor (IMR) PD-1 have efficacy in multiple indications, and molecules targeting other IMRs are in development. Increased understanding of IMR biology is required to design rational combination therapies and identify biomarkers of response and toxicity. Here we applied Single Cell Network Profiling (SCNP) to assess functional signaling across immune cell subsets in the context of IMR expression, using PBMC of CLL and healthy donors (HD).

Description of the technology Understanding a patients’ immune status not only from immune cell phenotyping, but also through analysis of functional signaling capacity, enables the generation of a more comprehensive understanding of the complex mechanisms responsible for immunological tolerance in cancer, and generates data that is complementary to other non-functional phenotypic data sets such...
متن کاملRecent Advances in T Cell Signaling in Aging
The immune system of mammalian organisms undergoes alterations that may account for an increased susceptibility to certain infections, autoimmune diseases, or malignancies. Well characterized are age related defect in T cell functions and cell mediated immunity. Although it is well established that the functional properties of T cells decrease with age, its biochemical and molecular nature is...
متن کاملThe Effect of SB431542, TGF-β Receptor Inhibitor, on HCV Replication in PBMCs of Patients with Chronic Hepatitis
Background and Aims: TGF-β is an effective cytokine in the viral replication cycle, which is also highly relevant to the pathogenesis of some viral infections. TGF-β induction by viral proteins is one of the ways to escape the virus from the immune system by inhibiting interferon signaling and other immune system factors. In recent years, the role of TGF-β and its inhibitor...
متن کاملFunctional Pathway Analysis Using SCNP of FLT3 Receptor Pathway Deregulation in AML Provides Prognostic Information Independent from Mutational Status
FMS-like tyrosine kinase 3 receptor (FLT3) internal tandem duplication (ITD) mutations result in constitutive activation of this receptor and have been shown to increase the risk of relapse in patients with acute myeloid leukemia (AML); however, substantial heterogeneity in clinical outcomes still exists within both the ITD mutated and unmutated AML subgroups, suggesting alternative mechanisms ...
متن کاملAnalysis of HLA-G Gene Expression in B-Lymphocytes from Chronic Lymphocytic Leukemia Patients
The human leukocyte antigen G (HLA-G) molecule exhibits limited tissue distribution, low polymorphism and alternative splicings that generate seven HLA-G isoforms. HLA-G exerts multiple immunoregulatory functions. Recent studies indicate an ectopic up-regulation in tumor cells that may favor their escape from anti-tumor immune responses. This study it is an effort to clarify the presence of HLA...
